Project Study: Abundance of Fungi and Bacteria in Salt Marsh Sediments (worktitle)

The investigation of capacity and rate of carbon sequestration in Wadden Sea salt marshes is part of the INTERFACE project. It is supposed that the process of biological decomposition plays a major role in the turnover of carbon in salt marsh sediments. Microorganisms are involved in the decomposition of biomass and can broadly be separated into the taxa bacteria and fungi. By means of quantitative polymerase chain reaction (Q-PCR), the abundance of major microorganism taxa can be determined (Smith & Osborn 2009). Together with data on vegetation structure and several abiotic site conditions the abundances of bacteria and fungi is planned to be included in a statistical evaluation.

The aim of this project study is to establish a method by which the abundances of bacteria and fungi can be determined in soil samples from salt marshes sediments.

Im Rahmen des INTERFACE-Projekts soll in einem Teilprojekt die Kohlenstoffbilanz von Wattenmeer-Salzmarschen untersucht werden. Es wird angenommen, dass biologische Zersetzungsprozesse eine bedeutende Rolle im Kohlenstoffumsatz in den Sedimenten von Salzmarschen spielen. Bei der Zersetzung von Biomasse sind Mikroorganismen (MO) beteiligt, die grob in die Taxa Bakterien und Pilze (Fungi) unterteilt werden können. Mit Hilfe der Methode Quantitative Polymerase Chain Reaction (Q-PCR) lassen sich die Abundanzen der beiden MO-Taxa bestimmen (Smith & Osborn 2009). Die Abundanzen sollen gemeinsam mit Vegetationsaufnahmen und verschiedenen Standortfaktoren in eine statistische Auswertung einfließen.

Ziel der Projektstudie ist es, die Methode zu etablieren mit der Bodenproben aus Salzmarschen auf die Abundanzen der MO-Taxa hin untersucht werden können.
