MOLECULAR CHARACTERIZATION OF ALTERNATIVE OXIDASE FROM CATHARANTHUS ROSEUS

Seiichiro Kiyota and Katsuhiro Sakano (National Institute of Agrobiological Resources, Tsukuba, Japan)

Alternative oxidase (AOX) is terminal oxidase of cyanide insensitive respiration. AOX is widely distributed among plants, fungi and protozoa. We studied role of AOX in biochemical pH-stat in plant cells. In the course of investigation, we isolated cDNA clone of AOX from cultured cells of Catharanthus roseus. This cDNA(1.4kb) is highly homologous to other plant AOXs. DNA bloting indicated that a single gene of AOX exist in Catharanthus roseus. Time course of AOX expression during culture under various conditions had been investigated.
